Abstract: The study on water quality and its impact on public health is among the most priority and contemporary issues in the field of environmental and health sciences. Water is an essential resource for the existence and development of life, while ensuring its quality remains one of the major global challenges. Increasing urbanization, industrial expansion, agricultural and livestock activities, together with climate change, have intensified pressure on aquatic environments, directly contributing to the deterioration of both surface and groundwater quality. In this context, the correlation between the physicochemical and microbiological parameters of water is an absolute necessity for environmental protection and public health safeguarding. The interest in conducting this study is related to the fact that the waters of the Buna river, Lake Shkodra, particularly in the Shiroka area and the Viluni Lagoon in Velipoja, hold significant importance for the local population, tourism, the natural environment, and the economic development of the region. These areas constitute aquatic systems with high ecological, touristic, and socio-economic value; however, they are simultaneously exposed to various anthropogenic and natural pressures. Urban and rural discharges, agricultural and livestock activities, the growth of tourism, as well as hydrological and climatic phenomena continuously affect water quality and contribute to the spread of microbiological degradation. This dissertation aims to analyze and interpret the correlation between the spatial variability of physicochemical parameters and microbiological indicators of water quality, with a particular focus on fecal coliforms, intestinal enterococci, and Escherichia coli. The study is based on laboratory analyses, international standard methods, and contemporary statistical techniques, which have enabled the identification of influencing factors on the distribution of pollution and on the microbiological quality of the studied waters. An important element of this study has been the identification of the relationship between microbiological contamination and its potential impact on public health. During the realization of this research, fieldwork, laboratory analyses, and statistical data processing were carried out. This study presents an innovation, as for the first time different ecosystems such as rivers, lakes, lagoons, and the sea have been analyzed across a wide geographical extent, contributing to the advancement of knowledge in the field of environmental analysis and water resource quality assessment. A total of 3934 laboratory analyses were performed, of which 3024 analyses correspond to physicochemical parameters, while 910 analyses were conducted for microbiological indicators. Through this study, the aim is to provide a scientific contribution to clarifying the processes that govern water pollution and to establish a solid basis for the sustainable management of water resources. The results of this
